specimens and using immunomagnetic beads (Miltenyi). Commercially available aortic medial SMC from six healthy donors were purchased from Promocell. Cells were used consistently at passage 3. We used two-dimensional electrophoresis with digital image analysis (SameSpots, Non-linear Dynamics) and tandem mass spectrometry to detect changes in proteome of atherosclerotic SMC. Results Analysis of 2D gel images revealed 29 proteins with a statistically significant difference in expression between medial and plaque SMC (p<0.05). Plaque SMC had decreased expression of mitochondrial protein ATP synthase subunit b but an increase in the oxidised form of peroxiredoxin-4, suggesting decreased mitochondrial function, possibly owing to oxidative stress. Glycolytic enzyme pyruvate kinase was also increased by 50% in plaque SMC. Furthermore, differences in protein expression between SMC from symptomatic and asymptomatic patients were also found. Plaque SMC from symptomatic patients exhibited increased expression of heat shock protein-60 and decreased levels of the antiinflammatory protein annexin I (p<0.05), compatible with a proinflammatory behaviour. These findings were confirmed by immunoblotting. Conclusions Our data demonstrate that plaque-derived SMC are exposed to higher levels of oxidative stress than control SMC. Differences between SMC from symptomatic and asymptomatic patients appear to reflect proinflammatory changes associated with plaque instability.

Augmented arterial wall expression of type-1 plasminogen activator inhibitor induced by thrombosis.

Type-1 plasminogen activator inhibitor (PAI-1), the primary physiological inhibitor of endogenous plasminogen activators, modulates fibrinolysis, cell migration, and tissue repair. To determine whether genetic expression of PAI-1 is augmented in the walls of vessels exposed to thrombi but not to a direct physical insult such as electrical injury, we induced arterial thrombosis in rabbit carotid...

Simvastatin suppresses dexamethasone-induced secretion of plasminogen activator inhibitor-1 in human bone marrow adipocytes

BACKGROUND Osteonecrosis of the femoral head is a common complication of high-dose glucocorticoid treatment. Intravascular thrombosis is thought to be associated with the ischemic state of the femoral head. Plasminogen activator inhibitor-1 (PAI-1) is an adipokine, which are physiologically active substances secreted from visceral and subcutaneous adipocytes. PAI-1 suppresses fibrinolysis by bi...

t-plasminogen activator inhibitor-1 polymorphism in idiopathic pulmonary arterial hypertension

AIM The aim of the present study was to identify the possible genotypic association of 3'UTR Hind III polymorphism of Plasminogen activator Inhibitor-1 (PAI-1) gene with idiopathic pulmonary arterial hypertension (IPAH). BACKGROUND IPAH is a disorder with abnormally raised mean pulmonary arterial pressure and increase in the resistance to blood flow in pulmonary artery. One of the pathologica...

Plasminogen activator inhibitor-1 messenger RNA expression is induced in rat hepatocytes in vivo by dexamethasone.

Plasminogen activator inhibitor-1 (PAI-1), the major physiologic inhibitor of tissue plasminogen activator (tPA), plays a crucial role in the regulation of fibrinolysis. Both hepatocytes and endothelial cells have been implicated as major sources of plasma PAI-1. To study the relative contribution of these cell types to hepatic PAI-1 production, we have separated hepatocytes and hepatic sinusoi...
